Tranmere's FA Cup campaign came to end at Prenton Park after Peterborough came from behind to claim a 2-1 victory.
Rovers took the lead in the first half through Shane Sherriff, but they were pegged back in injury time and Posh grabbed the winner midway through the second period.
The same team that defeated Brighton and Blackpool was due to start the game, but John Mullin limped out of the warm up and had to be replaced by Kevin Ellison, who was returning from injury. Taking Ellison's place on the bench was Craig Curran.
The starting formation against Peterborough
Rovers almost took the lead with a fine move in the fourth minute. Chris Shuker ran around his marker to collect a pass from John Thompson wide on the left, and the winger then made easy work of Guy Branston before crossing to Chris Greenacre at the near post. He met the ball well but his header was brilliantly turned over by Peterborough keeper Mark Tyler.
Ronnie Moore's side kept the pressure on, winning a couple of quick corners, before Shuker himself had a shot blocked after Greenacre and Steve Davies had combined well on the left wing.
Posh's first effort saw Danny Crow fire wide from 20 yards on 14 minutes, though the visitors had earlier tested the Rovers defence with a couple of deep swinging crosses.
At the other end Ellison shot over from his unusual central midfield position, while Gavin Ward produced a fine one handed save to keep out Richard Butcher's powerful right foot strike from 25 yards.
Appeals for handball on the edge of the Peterborough area were ignored by the referee on 29 minutes, but Rovers took the lead a minute later from the corner that followed.
Shuker floated the ball in and Shane Sherriff out jumped the visiting defence to head against the bar with the ball dropping over the line and into the net at the far post.
A drop of the shoulder put Thompson in space inside the Peterborough half on 36 minutes, but his shot from distance was blocked on the way to goal, while Ward saved low down from Adam Newton soon after.
Another low effort from outside the area from Crow was held by Ward on 39 minutes, but the action soon returned to the other end where Ellison smashed shot against the side netting from 16 yards after Greenacre's initial shot had been blocked.
With the half in stoppage time, Peterborough levelled. A quick attack saw Newton break free on the left, and his cross was turned in at the near post by Crow.
Rovers won a free kick in a central position right on the edge of the Peterborough area on 47 minutes for a foul on Ellison. Davies drilled in a low shot, but it struck the boot of a defender and went out for a corner.
The home side were the beneficiaries of some odd officiating on 53 minutes when, after first waving play on when Davies went down under a challenge near the half way line, the referee then changed his mind when spotting his assistant waving his flag on the touchline.
Davies and Ellison combined well on the left on 63 minutes, with the former seeing a low shot saved by the legs of Tyler.
At the other end Rovers survived a goalmouth scramble that saw Jude Sterling's shot go across the face of goal, and pressure eventually ended with a shot over the bar from Shane Huke.
Taylor saw dipping shot take a deflection and go just over the Peterborough goal on 67 minutes, but the visitors scored again with their next attack. A throw from the right bounced through the Rovers area and Crow nipped in from the back post to fire past Ward from close range.
Butcher fired over from 16 yards on 76 minutes and Rovers responded by a move to a 3-4-3 formation. Thompson pushed into midfield alongside McLaren with Calvin Zola coming on for Ellison.

Zola was denied an equaliser on 82 minutes by a fine save from Tyler, after the striker met Davies' cross on the edge of the six yard box.
